摘要
试验参考鲤鱼γ-2β干扰素(interferonγ-2β,IFNγ-2β)全长cDNA序列,在其两侧非翻译区和外显子设计2对引物,以提取的鲤鱼脾脏基因组DNA为模板进行PCR扩增,获得了IFNγ-2β的全长基因DNA序列。结果表明,获得的基因组DNA全长2084bp,GenBank登录号为JX181981,序列分析结果显示该基因进化较保守,和其他物种一样都含有4个外显子和3个内含子,各外显子碱基数与哺乳动物中相对应的外显子碱基数基本相同,且外显子、内含子剪接位点均遵守GT-AG规则。
The research aimed to carry out the cloning,identification and sequence analysis of genomic DNA of carp interferon γ-2β(IFNγ-2β).On the basis of the full-length cDNA of carp,we designed two pairs of primers in its bilateral untranslated regions and the exons to perform PCR that used the carp spleen genomic DNA as the template,the IFNγ-2β full-length genomic DNA sequence was obtained.The full-length genomic DNA was 2084 bp,the GenBank accession number was JX181981,the gene sequence analysis showed the evolution of conservative and contained 4 exons and 3 introns were like most of the other species,the number of bases of each exon closed to the mammalian exons correspondingly.The exon and intron splice sites were complied with GT-AG rules.
