摘要
运用 RAPD和 SSR技术 ,采用分离群体分组分析法 ( BSA)进行了小麦合成种 M53抗白粉病基因连锁的分子标记研究。结果表明 ,M53的抗白粉病基因由显性单基因控制 ,RAPD标记OPL0 9-170 0 与抗病基因连锁 ,遗传距离为 16.8c M。SSR标记 Xgwm2 0 5也与抗白粉病基因连锁 ,遗传距离为 9.3c M,通过 SSR标记将该基因定位于 5DS,标记与基因间的排列顺序为 :OPL 0 9-170 0 - 16.8c M-抗白粉病基因 - 9.3c M- Xgwm2 0
Random amplified polymorphic DNA (RAPD) and simple sequence repeats (SSR) were employed to detect molecular markers linked to powdery mildew resistance gene in Triticum durum Aegilops squarrosa amphidiploid M53. The resistance was controlled by single dominant gene. A RAPD marker OPL09 -1700 was proved to link to the resistance gene with genetic distance of 16.8 cM. A SSR marker Xgwm205 was also proved to link to the resistance gene and the genetic distance is 9.3 cM. This powdery mildew resistance gene has been located in 5DS and the sequence in 5DS is OPL09 -1700 Pm Xgwm205.
