摘要
以BJ-299、Tx622B、W456、忻粱52和Roma 5个高粱[Sorghum bicolor(L.)Moench]品种的基因组DNA为模板进行PCR扩增以筛选SSR分子标记引物。设计不同的退火温度和扩增程序,对250对SSR引物进行筛选和反应程序的优化。结果表明,250对SSR引物中有223对引物扩增成功,大部分引物的退火温度为55~57℃,共筛选出125对在耐盐碱品种BJ-299与盐碱敏感品种Tx622B间表现出多态性的SSR引物。
Genomic DNA of 5 Sorghum bicolor varieties,BJ-299,Tx622B,W456,Xinliang 52 and Roma,were used as template for screening SSR primers.By adjusting the annealing temperature and amplification procedure,223 out of 250 pairs of primers were successfully amplified,of which the annealing temperature was mostly 55~57 ℃.125 SSR markers showed polymorphism between saline alkali resistant variety BJ-299 and saline alkali sensitive variety Tx622B.
