静态负荷所致肌肉损伤的能量代谢变化 被引量:8

摘要 目的 通过观察静态负荷过程中家兔骨骼肌能量代谢的变化 ,探讨静态负荷致肌肉损伤的机制。方法 应用荧光法测定三磷酸腺苷 (ATP)及肌酸磷酸 (PCr)的含量 ,应用组织化学方法和分光光度法测定糖原的含量 ,应用生物化学方法测定血清中肌酸激酶 (CK)的含量。结果 在实验的第 1、2、4天 ,观察组家兔腰肌ATP含量有不同程度的降低 ,实验的第 6天 ,ATP含量明显下降 [(4.18±0 .2 5 ) μmol/g],明显低于对照组 [(4.5 5± 0 .18) μmol/g],差异有显著性 (P <0 .0 5 )。观察组家兔腰肌PCr含量呈现逐渐下降趋势 ,实验的第 4、6天 ,实验组PCr含量 [(14.17± 5 .98)、(10 .83± 4.99) μmol/g]与对照组 [(2 3.6 7± 5 .92 )、(2 5 .0 0± 5 .48) μmol/g]比较 ,差异有显著性 (P <0 .0 5、P <0 .0 1)。随着实验时间的延长 ,糖原含量下降 ,实验的第 6天 ,观察组糖原含量 [(0 .2 2± 0 .0 7)g/ 10 0g]明显低于对照组[(0 .33± 0 .0 6 )g/ 10 0g],差异有显著性 (P <0 .0 5 )。组织化学方法观察糖原变化结果与此一致。观察组血清CK活力在实验的第 1天 [(5 5 2± 38)U/L]即明显高于对照组 [(2 5 8± 2 8)U/L],差异有显著性(P <0 .0 1)。结论 施加静态负荷后 ,骨骼肌细胞能量代谢发生障碍 ,可能是造成肌肉损伤的原因之一? Objective To study the mechanism of muscle injury caused by static load through observing the changes of energy metabolism in rabbit muscle under static load. Methods Rabbits were exerted difinite static load on their lumbar region to induce muscle injury.ATP and creatinephosphate(PCr) were measured by means of fluorescence spectrophotometry.Glycogen was measured by using spectrometry and histochemistry methods respectively.Biochemistry method was applied to measure the activity of serum CK. Results ATP contents of skeletal muscle in experimental group became lower than those in control group on the 1st,2nd,4th d of the experiment and there was significant difference between two groups[(4.18±0.25) vs (4.55±0.18)μmol/g]( P <0.05) on the sixth day.The contents of PCr in experimental group decreased obviously on the 4th d and 6th d[(14.17±5.98),(10.83±4.99)μmol/g respectively] than those in control group[(23.67±5.92),(25.00±5.48)μmol/g respectively, P <0.05, P <0.01].Glycogen decreased with prolongation of experimental time and there was significant difference between two groups on the 6th d[(0.33±0.06) g/100 g vs (0.22±0.07)g/100 g]( P <0.05).The same changes of glycogen were observed by using histochemistry method.There had been significant difference in creatine kinase activity between experimental group[(552±38)U/L] and control group[(258±28)U/L]( P <0.01) since the 1st d. Conclusions Static load could lead to the disturbance of energy metabolism in muscle and it may be one of the reasons for muscle injury caused by static load.
