摘要
目的简化分离、纯化核心蛋白聚糖的程序 ,了解其对瘤细胞生长的抑制作用。方法经匀浆、溶解沉淀、透析、研磨等步骤制备核心蛋白聚糖 ,MTT染料结合法检测A5 49、BT32 5等四种癌细胞体外生物学活性。结果简化法制备的核心蛋白聚糖得率为 3mg/g大鼠肌肉 ,SDS PAGE显示在相对分子质量 36~ 38kD间有两条带 ,除明显抑制A5 49生长外 ,对Hela、胃 790 1及BT32 5三者同样具有抑制生长活性 ,抑制率依次为 6 9.19%、48.19%、5 4.47%、6 7.80 %。结论此法成本低、纯度好。
PurposeThe aim is to simplify the isolation and purification of Decorin and understand the bioactivity of suppressing other cancer cell growth. MethodsDecorin is prepared from rat muscle mainly by homogenizing, dissolving precipitation,dialyzing as well as grinding. Assay the bioactivity of 4 kinds of cancer cells in vitro, such as A549, BT325, etc, by the method of MTT(dye combination).ResultsThe content of Decorin prepared by the simplified method has increased by 50%. About 36~38 kD stained band was observed on SDS PAGE. Besides A549, decorin suppressed proliferation of Hela, Stomach, BT325, A549 cells. The percentage of inhibition is 69.19%, 48.19%, 54.47%,67.80% respectively. ConclusionThis simplified method not only is of low cost but also has a higher purity. Both productivity and bioactivity of Decorin are increased significantly.
