摘要
目的 :研究微量元素硒诱导人类白血病细胞凋亡的作用及其相关的基因调控。方法 :体外培养人急性早幼粒白血病细胞 (HL 60 ) ,加入硒使其终浓度分别为 0、0 .1 μg/ml、1 μg/ml、2 μg/ml,作用 4 8h后收集细胞 ,分别作形态学观察、DNA琼脂糖凝胶电泳、流式细胞光度计分析、TUNEL及bcl 2 /bax基因蛋白表达的细胞化学检测。结果 :上述多项指标的分析结果均证明元素硒能诱导癌细胞凋亡。通过流式细胞光度计检测到对照组、0 .1 μg/ml、1 μg/ml、2 μg/ml加硒组的凋亡百分率分别为 4 .4 3 %、8.3 6%、3 7.91 %、63 .4 7%。这说明浓度为0 .1 μg/ml的硒没有明显的诱导癌细胞凋亡的作用 ,而浓度为 1 μg/ml、2 μg/ml时凋亡细胞显著增多并呈剂量依赖关系。随着凋亡细胞的增多 ,bcl 2基因表达相应减弱 ,而bax基因表达相应增强。结论 :微量元素硒可诱导HL 60细胞凋亡 ,并且呈剂量依赖关系 ,凋亡的发生受bcl 2
Objective:To study the apoptotic induction effect of trace element selenium on human acute promyelocytic leukemic(HL 60)cells and the regulation of related genes.Methods:HL 60 cells were cultured in vitro, selenium was added into cell medium.The final concentrations of selenium were 0,0.1,1 and 2 μg/ml.After 48 hours incubation, cells were harvested for morphologic observations under the light microscope, flow cytometric analysis (FCM),DNA agarose gel electrophoresis, TdT mediated dUTP nick end labeling (TUNEL) and cytochemical analysis for the level of bcl 2 and bax gene expression in the cells.Results:The apoptotic rates by FCM in control, 0.1,1 and 2 groups were 4.43%,8.36%,37.91% and 63.47%, respectively. The data indicated that in group 0.1μg/ml, apoptosis induction effects of selenium is not obvious,while in group 1 μg/ml and 2 μg/ml, this effect was significantly increased and dose dependent. The results of DNA agarose gel electrophoresis, TUNEL and cytochemical analysis showed corresponding presentations among these four groups.Along with the increase of number of apoptotic cells,the expression of bcl 2 genes reduced and that of bax genes enhanced.Conclusion:Trace element selenium could induce apoptosis of HL 60 cell.This effect is dose dependent.The apoptosis induction was related to the expression of bcl 2/bax genes.
