低氧性肺动脉高压大鼠血清IGF-1的变化 被引量:1

摘要 目的 探讨慢性低氧大鼠血清中胰岛素样生长因子 1(IGF 1)浓度、肺小动脉壁IGF 1多肽表达的变化及其与低氧性肺动脉高压 (HPH)之间的关系。方法 用放射免疫分析法检测大鼠血清中IGF 1浓度 ;用免疫组织化学染色法观察肺小动脉壁IGF 1多肽的表达 ;用图像分析技术观察肺小动脉的形态改变和IGF 1多肽的表达强度。结果 低氧性肺动脉高压大鼠管壁增厚 ,管腔狭窄 ,反映管壁增厚的两个指标MT %和MA %均显著高于正常对照组 (P <0 0 5 ) ;灰度扫描示低氧大鼠肺小动脉壁的阳性程度显著大于正常对照组 (P <0 0 1)。低氧大鼠血清IGF 1水平明显高于正常对照组 (P <0 0 5 ) ;大鼠血清IGF 1水平与平均肺动脉压 (mPAP)呈显著正相关 (r =0 745 ,P =0 0 0 2 )。结论 在低氧性肺血管重建的发生机制中 ,IGF 1不仅以自分泌和旁分泌的方式刺激血管平滑肌细胞增生肥厚 ,而且还存在内分泌机制。 Objective To study the change of IGF 1 concentration in serum,of IGF 1 polypeptide in the walls of pulmonary arterioles,and the relationship with hypoxic pulmonary hypertension.Methods The concentration of IGF 1 in the rat serum was measured by radioimmunoassay.The change of expression of IGF 1 polypeptide was observed by immunohistochemical staining on the walls of pulmonary arterioles.Morphological change and the level of IGF 1 polypeptide in pulmonary arterioles were determined by image pattern analysis technique.Results Compared with the normal control group,the walls of pulmonary arterioles in the rats with hypoxic pulmonary hypertension were observed to be thickened with both MT% and MA% higher significantly than those of normal ( P <0 05).The positive degree of gray scale scanning in the pulmonary arterioles in the hypoxic rats was higher than that in the normal rats.The level of IGF 1 in serum in the hypoxic rats was higher than that in the normal rats ( P <0 05).The level of IGF 1 in the rat serum was positively correlated with mPAP ( r =0 745,P=0 002).Conclusion In the pathogenesis of hypoxic pulmonary vascular remodelling,IGF 1 stimulate vascular smooth muscular cell proliferation not only in autocrine and paracrine manner,but also in the endocrine manner. Key words Hypoxic pulmonary hypertension IGF 1 polypeptide IGF 1 level in serum Endocrine manneryan,LI Shao qun.Sanitary and Anti epidemic Station of Guangzhou City,510080,China.Abstract:In order to strengthen hygienic supervise of detergent in the market,we engaged in toxic studying on welson gleam detergent in NIH mice.Results indicated that it is in a low toxicity,acute toxicity median lethal dose(LD 50 ) is 2450mg/kg.It didnt increase that the rate of sperm shape abnormality and the contents of micronucleus test of bone marrow cell.White blood cell(WBC) was decreased as the dose went up in thirty days feeding tests,and the differences were signifficant between tests groups and control group( P <0 01).Other blood biochemical index changed all in normal range.
