摘要
目的 研究外源性三磷酸腺苷 (ATP)在大鼠肝脏低温保存中的作用及其机制。方法 采用持续低温机器灌流保存模型 ,利用高效液相色谱、光镜及放射自显影等技术测定低温保存大鼠肝脏的能量代谢、功能、形态学变化及外源性ATP是否能进入肝细胞内。结果 灌流液中不含ATP及Mg Cl2 组 (A组 )和灌流液中含 5mmol/LATP、不含MgCl2 组 (B组 ) ,随保存时间的延长 ,大鼠肝细胞内ATP及Atkinson’s能量转换 (EC)迅速下降 ,两组差异不显著 (P >0 .0 5 ) ,而灌流液中含 5mmol/LATP及 5mmol/LMgCl2 组 (C组 ) ,ATP及EC则下降缓慢 (P <0 .0 1) ;A、B组乳酸脱氢酶 (LDH)及天冬氨酸转氨酶 (AST)迅速增高 (P >0 .0 5 ) ,C组上升较A、B组缓慢 (P <0 .0 5 ) ;保存 2 4h后A组、B组形态学变化基本一致 ,均可见肝细胞肿胀明显 ,C组则显示肝细胞水肿较轻 ;放射自显影显示 [α - 32 p]ATP进入肝细胞内。结论 外源性ATP在大鼠肝脏低温保存中有保护作用 ,但需Mg2 +参与 ;外源性ATP能够进入低温保存的大鼠肝细胞内。
Objectives To investigate the protective effect of exogenous adenosine triphosphate (ATP) on rat liver during cold storage and its mechanism.Methods Using continuous hypothermic machine perfusion model, nucleotides in hepatocytes were determined with HPLC, activities of LDH & AST in the perfusate measured, and histological changes observed in different groups.[α- 32p]ATP was used to clarify the underlying mechanism of protective effect of ATP.Results The intracellular levels of ATP and EC were decreased rapidly after hypothermic preservation in the group A (none of ATP and MgCl 2 in the perfusate ) and group B (5?mmol/L ATP and no MgCl 2 in the perfusate), while a higher ATP, EC level and slower decreasing rate were observed in the group C (both 5?mmol/L ATP and 5?mmol/L MgCl 2 in the perfusate) (P< 0.01). As compared with groups A and group B, the activities of LDH and AST in the group C were lower (P< 0.05). More severe hepatocyte damage appeared in the group A and group B than in the group C. Radioactive [α- 32p]ATP was found to enter the cold stored rat hepatocytes.Conclusions Exogenous ATP showed protective effects on rat liver during cold storage, Mg 2+ was indispensable. It was observed that exogenous ATP could enter cold stored rat hepatocytes.
