摘要
目的 :研究烧伤后大鼠骨骼肌组织泛素蛋白酶体 (ubiquitin proteasome)蛋白降解途径组成成分中泛素表达的变化 ,探讨烧伤后骨骼肌蛋白降解的增强与泛素蛋白酶体蛋白降解途径表达变化间的关系。方法 :借助烫伤大鼠动物模型 ,利用骨骼肌离体孵育系统 ,应用氨基酸自动分析仪测定孵育液和骨骼肌组织中酪氨酸(Tyr)和 3甲基组氨酸 (3MH)量的变化 ,用 RNA印迹技术 (Northern blot)方法测定骨骼肌组织中泛素m RNA表达水平。结果 :烧伤后大鼠骨骼肌释放 Tyr和 3MH增多 ,以 3MH增加显著 ;烧伤后骨骼肌组织中泛素 2 .4kb的 m RNA表达上调明显。泛素 m RNA表达增加量与 Tyr和 3MH增加量间均呈显著正相关。结论 :烧伤后大鼠骨骼肌蛋白降解明显增强 。
Objective:To study the change in proteolytic rate in rat skeletal muscle after burn injury.Also,we observe the change in ubiquitin mRNA expression of ubiquitinproteasome pathway at different time points in rat skeletal muscle,and to investigate the relationship between proteolytic rate of rat skeletal muscle and ubiquitinproteasome pathway.Methods:With the help of rat model of burn injury,we determined the content of 3methylhistidine(3MH) and tyrosine (Tyr) in muscle incubation system in vitro by amino acid automatic analyser and the level of ubiquitin mRNA expression in different types of skeletal muscle by northern blot analysis.Results:The concentrations of 3MH and Tyr in incubation medium rat of extensor digitorium logus (EDL) muscle increased apparently.The level of ubiquitin mRNA expression in EDL muscle increased markedly after burn injury in rats compared with control group.There was a positive correlation between the expression of ubiquitin mRNA and respective contents of Tyr and 3MH.Conclusions:Proteolytic enhancement of rat's skeletal muscle occurred after burn injury.The enhancement of skeletal muscle proteolysis in burned rats was attributed to activation of ubiquitinproteasome pathway in transcriptional level.
