摘要
目的 :观察烫伤对大鼠血浆肿瘤坏死因子 α(TNFα)水平的变化及 α 黑色素细胞刺激激素(α MSH)和 KPV合成肽的调节作用。方法 :用酶联免疫吸附 (EL ISA )法检测正常对照组、轻度烫伤组和重度烫伤组大鼠血浆 TNFα的水平 ,用烫伤前大鼠尾静脉注射 TNFα和白介素 1β(IL 1β)中和抗体及腹腔注射α MSH和 KPV等调节重度烫伤大鼠血浆 TNFα的水平。结果 :与正常对照组血浆 TNFα水平相比 ,烫伤后 12小时 ,轻度烫伤组大鼠血浆 TNFα水平显著升高 (P<0 .0 1) ;与轻度烫伤组相比 ,重度烫伤组大鼠血浆 TNFα水平升高更加明显 (P<0 .0 1)。体内注射 TNFα和 IL 1β中和抗体及 α MSH和 KPV均能明显减轻重度烫伤组大鼠血浆高 TNFα血症状态。结论 :烫伤可使大鼠血浆 TNFα的水平明显升高 ;体内注射TNFα、IL 1β中和抗体及 α MSH和 KPV均可在一定程度上减轻重度烫伤所致的血浆
Objective:To observe the changes in plasma tumor necrosis factorα(TNFα) levels in scalded rats and the regulatory effect of αmelanocytestimulating hormone(αMSH) and KPV peptide on its level.Methods:Plasma TNFα level in normal,mildly scalded and severely scalded rats was determined with enzymelinked immunosorbent assay (ELISA).Plasma TNFα concentrations were regulated by intravanous injection of antirat TNFα or IL1β antibodies,and intraperitoneal administration of αMSH and KPV peptide (AcDLysLProDVal) before scald.Results:TNFα level in rats after mildy scald was higher than that in control group 〔(48 436±10 327)ng/L vs.(9 937±13 284)ng/L, P <0 01〕.It was significantly increased in severely scalded rats (111 177±23 973)ng/L as compared with that in mildly scalded rats ( P <0 01).The injection of antirat TNFα and IL1β antibodies,αMSH and KPV attenuated the increase of TNFα levels in severely scalded rats.Conclusions:There is a significant increase in plasma TNFα level in rats after scald.The administration of antirat TNFα and IL1β antibodies,or αMSH and KPV might attenuate this increase.
