摘要
目的:探讨烧伤后早期腹腔巨噬细胞表达磷酸化酪氨酸(Tyr P)、核因子 Kappa B(NF κB)m RNA 及分泌细胞因子的动态变化。方法:采用小鼠20% 体表面积的Ⅲ度烫伤模型,分离和培养小鼠腹腔巨噬细胞。用免疫组织化学方法观察烫伤后24 小时内不同时间Tyr P的变化;用逆转录聚合酶链反应(RTPCR)方法观察NF κBm RNA表达的情况;用酶联免疫吸附(ELISA)方法测定上述细胞培养上清中肿瘤坏死因子 α(TNF α)、白介素 6(IL 6)的水平。结果:烫伤后1 小时腹腔巨噬细胞的Tyr P表达就明显增高,与假烫伤组比较差异显著(P< 0.01),这种改变在伤后24 小时内持续存在;烫伤后1 小时NF κBm RNA 表达明显增强,以伤后2 小时为明显(P< 0.01),伤后4、6 小时表达逐步减弱,但仍较假烫伤组高(P均< 0.05);烫伤小鼠腹腔巨噬细胞培养上清中TNF α和IL 6 水平明显增高,烫伤后其含量在不同时期有所不同,与假烫伤组比较差异显著(P均< 0.01)。烫伤小鼠腹腔巨噬细胞Tyr P、NF κBm RNA 与TNF α、IL 6 呈正相关。结论:烫伤可使巨噬细胞中Tyr P、NF κBm RNA 表达增强,分?
Objective:To observe the early dynamic changes in tyrosine phosphorylation (TyrP),nuclear factorκB (NFκB) mRNA and secretion of cytokines in peritoneal macrophages in severely burned mice.Methods:BALB/C mice were subjected to a 20 percent fullthickness scald injury.The peritoneal macrophages in different intervals postburn were obtained to measure TyrP and NFκB mRNA expression by immunohistochemical and reverse transcriptionpolymerase chain reaction analysis.The levels of tumor necrosis factorα(TNFα) and interleukin6(IL6) in the medium were determined by ELISA.Results:Significantly increased expression of TyrP at different intervals postburn was found,and NFκB mRNA expression markedly increased at 1 hour,peaking at 2 hours.The concentrations of TNFα and IL6 produced by macrophages were obviously higher in burned mice than in control mice.Positive correlations were found between TyrP,NFκB mRNA expression and TNFα,IL6 secretion.Conclusions:In the macrophages of the burned mice,the expression of TyrP and NFκB mRNA and the secretion of TNFα and IL6 were increased obviously.These changes may play an important role in mediating multiple organ damage by bacterial endotoxin.
