不同材料对人骨髓间充质干细胞增殖的影响 被引量:10

The effect of different vehicles on the proliferation of bone marrow mesenchymal stem cells

摘要 目的 了解冻干骨和羟基磷灰石对骨髓间充质干细胞增殖的影响 ,为细胞移植载体的选择提供依据。方法 人骨髓间充质干细胞分成 3组 ,分别用冻干骨提取液、羟基磷灰石提取液和普通培养液进行培养对照。在培养 2 4h、2 8h后用流式细胞仪分别对 3组细胞进行细胞周期检测。结果  3组细胞均未出现DNA异常倍体。 2 4h时 ,在整个增殖期、DNA合成期、有丝分裂期 ,3组细胞之间差异均有显著性。其中 ,冻干骨主要抑制了细胞的DNA合成 (冻干骨组均值为 2 17% ,对照组均值为18 98% ) ;羟基磷灰石主要抑制了细胞的有丝分裂 (羟基磷灰石组均值为 6 69% ,对照组均值为12 0 2 % )。但到 2 8h ,各组在DNA合成期、有丝分裂期的细胞数量趋于接近。结论 两种材料对骨髓间充质干细胞的增殖均有短暂的抑制作用 。 Objective To investigate the effect of the freeze dried bone and hydroxyapatite on the proliferation of bone marrow mesenchymal stem cells. Methods Human bone marrow mesenchymal stem cells were divided into three groups. One was cultured with the extract of freeze dried bone, the other one with the extract of hydroxyapatite, and the last one with control medium (10% FBS DMEM). After 24 and 28 h, the cells were detected for their cycle parameters with flow cytometer. Results No abnormal ploidy was found in three groups at two time points. The amounts of the cells, which were at multiplication phase, DNA synthesis phase, and mitosis phase, were different significantly among three groups after 24 h culture. The freeze dried bone mainly inhibited the DNA synthesis, and the hydroxyapatite mainly inhibited the mitosis. But till 28 h, the cell number of three groups at DNA synthesis and mitosis phases tended to be close with one another. Conclusion Although the two materials can inhibit the proliferation of mesenchymal stem cells for a short period of time, the inhibitory action will disappear gradually along with time and change of circumstance, but they both can be used as vehicle of bone marrow mesenchymal stem cells.
