新型肾综合征出血热EIA诊断试剂盒的研制 被引量:1

摘要 用多肽合成仪合成汉坦病毒核衣壳蛋白 (NP) (aa1 7~ 6 6 ) 5 0个氨基酸残基的核心序列 ,以此为抗原 ,建立特异性检测抗汉坦病毒IgG和IgM的酶联免疫吸附剂测定法 (ELISA) ,通过实验发现这一序列具有较强的抗原性 .研制出了以合成肽为抗原的新型特异性检测抗汉坦病毒IgG和IgM的诊断试剂盒 .通过对肾综合征出血热抗体阳性血清的检测 ,效果良好 . Hantaviruses (HV), belonging to the family Bunyaviridae, cause hemorrhagic fever with syndrome (HFRS). HFRS is characterized by fever, renal dysfunction and bleeding complications. Small mammals, the mainly rodents, are the natural reservoirs. In order to establish a new specific serological diagnostic kit to test HFRS. The aa17-66 of NP epitope has been synthesized by solid phase peptide synthesis .Then the synthetic peptide is applied as an antigen to test the specific antibody (IgG,IgM) by EIA. The synthetic peptide shows stronger high antigenic. The anti-HV IgM and IgG are detected specially to develop a specific EIA kit for the diagnosis of HFRS.
