雷公藤内酯醇对人树突状细胞体外发育及免疫学功能的影响 被引量:35

摘要 目的 :观察雷公藤内酯醇对人树突状细胞 (dendriticcells ,DC)体外发育及免疫学功能的影响。  方法 :人DC体外培养时 ,加入不同剂量 (0、0 3、1 0、3 0、10、30、10 0 μg/L)的雷公藤内酯醇处理 ,观察DC的生成情况 ,以流式细胞仪分析各处理组生成细胞的免疫表型及抗原内吞能力 ,体外混合淋巴细胞反应检测各组细胞的抗原呈递功能 ,TUNEL方法检测细胞凋亡。  结果 :体外培养 7天后 ,10 μg/L及以上剂量雷公藤内酯醇处理组细胞大部分破碎死亡。低剂量 (0 3、1 0、3 0 μg/L)雷公藤内酯醇处理对生成的DC的形态及数量、共刺激分子的表达、内吞蛋白质抗原OVA FITC的能力、同种T细胞的免疫刺激活性均无显著影响 ;但正常DC与同种T细胞混合培养时 ,雷公藤内酯醇以剂量依赖性方式抑制T细胞的增生 ;正常DC以 30 μg/L雷公藤内酯醇处理 2 4h ,有 5 4%的细胞发生凋亡 ;  结论 :低剂量 (0 3、1 0、3 0 μg/L)的雷公藤内酯醇处理对DC的体外发育及免疫功能均无显著影响 。 Objective:To study the effects of triptolide on cell growth and immuologic functions of human dendritic cells(DC) in vitro. Methodology:Cultured DC was treated with different dosage of triptolide (0,0 3,1 0,3 0,10,30 and 100 μg/L). The immunologic phenotype and antigen endocytosis activity of the DC were analyzed by flow cytometry.T cell stimulatingactivity was determined by allo MLR.The apoptosis was detected by terminal deoxynucleotidyl transferase mediated dUTP biotin nick end labeling(TUNEL). Results:High dose of triptolide induced all cells death on day 7.However,the DC treated with low dose of triptolide(0 3,1 0,and 3 0 μg/L)showed no significant changes on the cell shape,cell number,co stimulation molecular expression,antigen endocytosis activity and allogeneic T cells stimulating activity.The proliferation of T cells was inhibited by triptolide by dose dependent way when normal DC was co cultured with allogeneic T cells.When DC was treated with triptolide with the dosage of 30 μg/L, there were about 54% of apoptosis induced in normal DC. Conclusion:Triptolide in lower dose has no significant effect on cell growth and to the immuologic function of DC,however,tripdide in higher dose can inhibit DC growth and induce DC apoptosis in vitro.
