亚低温复合灯盏花素对沙土鼠脑缺血再灌注损伤的影响

Effects of hypothermia combined with erigeron on reperfusion injury following forebrain ischemia in gerbils

摘要 目的 研究亚低温复合灯盏花素对脑缺血后沙土鼠病理学和行为学的影响。方法 采用沙土鼠前脑缺血模型 ,缺血时间 10min。动物分为假手术组、常温组、亚低温组、灯盏花素组和亚低温复合灯盏花素组。分别于再灌注第 2天和第 4天进行组织病理学和开阔法行为学检查。结果 组织病理学结果显示 ,再灌注第 4天海马CA1区存活神经元计数亚低温组或灯盏花素组较常温组明显增多 (P <0 .0 1) ,而与亚低温复合灯盏花素组相比则明显减少 (P <0 .0 1)。行为学检查结果显示 ,常温组沙土鼠的探索活动较假手术组明显活跃 (P <0 .0 1) ,亚低温组的探索活动较常温组减弱 (P <0 .0 5 ) ,亚低温复合灯盏花素组较亚低温组或灯盏花素组减弱 (P <0 .0 5 )。结论 亚低温复合灯盏花素能减轻脑缺血后海马CA1区锥体细胞延迟性死亡和动物行为学障碍。 Objective To study the effects of hypothermia combined with erigeron breviscapus on histopathology and behavior following forebrain ischemia in gerbils. Methods The ischemia was induced by occlusion of the two common cartoid arteries for 10 min. The animals were randomly divided into sham-operated (Sham) group, normothermia (NT) group, mild hypothermia (HT) group, erigeron breviscapus (EB) group and mild hypothermia combined with erigeron breviscapus (HE) group. Histopathology examination and open field test were carried out on the second day and the fourth day. Results Histopathology on the fourth day revealed the number of survival neurons in the hippocampal CA1 area was larger in the HT and EB groups than that in the NT group (P<0.01) and smaller than that in the HE group (P<0.01). The exploring activity of gerbils in HT group was less than that in NT group (P<0.05), the exploring activity in HE group was less than that in either the HT or the EB group (P<0.05). Conclusion Hypothermia combined with erigeron breviscapus attenuates the delayed death of neurons in hippocampal CA1 area as well as the behavior deficits in gerbils after transient forebrain ischemia.
