摘要
从雷公藤 (TripterygiumwilfordiiHook .F .)根心部分的乙醇提取液中 ,采用硅胶柱色谱法和TLC方法得到纯的雷公藤内酯甲 (wilforlideA) .用IR ,1HNMR和元素分析表征 .分子为五环三萜γ—内酯 .从AM 1计算结果可看出 ,分子中C(2 2 ) -C(2 3)为烯键 ,C(11)连有羟基 ,C(33) -O(3)为羰基 ,整个分子呈起伏状 ,不在一个平面 .仲羟基中的O(1)及γ—内酯中的O(3)原子净电荷大 ,使得分子极性也大 ,易在O(1) ,O(3)部位发生亲核反应、氧化还原反应 。
By silica gel column chromatography and TLC method,a pure wilforlide A was separated from the ethanolic extract of tripterygium wilfordii Hook.F. root.The compound was characterized by IR, 1HNMR and elemental analysis. The title compound is a five-ringed terpene, in which, the atoms, C(22) and C(23), are doublly bonded, the hydroxy group attaches to C(11) and, C(33) and O(3) form a carbonyl functionality. Whole skeleton of molecule stretches folded. The result of AM1 calculation shows that the larger charge densities on O(1) and O(3) of γ-lactone makes the molecule polar and reactive to nucleophilic and redox reaction.
