摘要
用 PCR方法从禽白血病病毒 ( avian leukosis virus,ALV)亚群毒株 RAV-1、RAV-2感染或未感染的SPF鸡胚成纤维细胞 ( CEF)分别扩增出 1 .2 kb基因片段。 RAV-1囊膜基因可被 Bgl 分为 2条相近的 60 0bp片段 ,RAV-2囊膜基因被 Bam H 分为 2条约 60 0 bp片段 ,对照组基因片段 (内源性病毒 )不被 Bgl 和Bam H 切割。结果显示 ,ALV囊膜基因的 PCR/RFLP可用于诊断禽白血病和鉴别不同的 ALV毒株。
A 1.2 kb PCR fragment was obtained from SPF chicken embryo fibroblast cell(CEF),which was infected with avian leukosis virus(ALV) RAV 1,RAV 2 or not with any ALV as control.The envolope gene fragment of RAV 1 was digested into two similar size fragments of 600 bp with BglⅡ and that of RAV 2 was digested into two similar size fragments of 600 bp with BamHⅠ as well,whereas the fragment from no infected CEF(endogenous virus) wasn′t digested with BglⅡ or BamHⅠ.The PCR/RFLP of ALV envolope gene was proved to be able to diagnose and identificate different ALV strains.
