摘要
目的探讨活动期SLE患者外周血中T、B细胞表面Fas和bcl-2的表达水平及其与T、B细胞凋亡的关系。方法采用流式细胞术,测定活动期SLE患者外周血T、B细胞表面Fas和bcl-2的表达,并同时测定患者血中T、B细胞的凋亡。结果①活动期SLE患者外周血中B细胞及CD8+T细胞表面bcl-2的表达明显高于正常人(分别为P∨0.05和P∨0.01);CD4+及CD8+T细胞表面Fas的表达高于正常人(分别为P∨0.05和P∨0.01);B细胞表面Fas的表达降低(P∨0.01);CD4+T细胞bcl-2的表达下降(P∨0.01)。②活动期SLE患者血中B细胞的凋亡率明显下降,CD8+T细胞数较正常对照组增加,CD4+T细胞低于常人(分别为:P∨0.01、P∨0.05和P∨0.01)。结论SLE患者体内淋巴细胞凋亡的异常与T、B细胞表面Fas和bcl-2基因表达的异常有关。
Aim To explore Fas and bcl 2 gene expressions on T and B lymphocytes in peripheral blood from active SLE patients and their relationship to lymphocytic apoptosis. Methods Expressions of Fas and bcl 2 genes on T and B lymphocytes and apoptotic cells in peripheral blood from the patients and control group were determined by flow cytometer. Results As compared with control group,the expressions of bcl 2 gene on B cells and CD8+T cells were increased significantly(P∨0.05 and P∨0.01 respectively), while decreased in CD4+ T cells (P∨0.01). Fas gene expressions on CD4+ and CD8+ T cells were increased (P∨0.05 and P∨0.01, respectively) and decreased in B cells(P∨0.05). The percentage of apoptotic B cells in SLE group was more than that in control group (P∨0.05). Conclusion Abnormal expressions of Fas and bcl 2 genes on T and B cells were related to lymphocytic apoptosis.
