有丝分裂激活蛋白激酶在人前列腺癌细胞中的激活 被引量:4

Activation of mitogen activated protein kinase(MAPK) in human prostate carcinoma cells

摘要 目的 探讨前列腺癌 (PCa)细胞中有丝分裂激活蛋白激酶 (MAPK)的激活。 方法 采用免疫沉淀和Westernblot方法 ,检测人PCa细胞株LNCaP及DU 145中磷酸化MAPK的表达及雄激素或表皮生长因子 (EGF)对其表达的影响。 结果 在DU 145中 ,磷酸化MAPK表达本底水平比LNCaP高 10倍。雄激素和EGF处理后 8hLNCaP中MAPK激活水平升高 ,2 4h达最高 ,分别比本底高约 10倍和 5倍 ,在DU 145中仅EGF可促使MAPK激活水平升高 ,8h开始升高 ,2 4h达最高 ,比本底高约 10倍。 结论 在雄激素非依赖性人PCa细胞增生中 ,MAPK激活比在雄激素依赖性人PCa细胞中作用大 ,MAPK激活也参与了雄激素和EGF的增生刺激作用。 Objective To investigate the activation of MAPK in prostate carcinoma cells. Me- thods In human prostate carcinoma cell lines DU-145 and LNCaP, the phosphorylation of MAPK was evaluated by using immunoprecipitation and Western blot analysis and the effect of EGF or androgen on the MAPK activation was also measured. Results The basal level of activation of MAPK was 10 folds higher in DU-145 than in LNCaP. Both androgen and EGF could stimulate the activation of MAPK in LNCaP cells, while only EGF rather than androgen could activate the MAPK in DU-145. Up-regulation of phosphorylated MAPK was found after 8h of treatment by either androgen or EGF, and reached the highest le- vel after 24h which was 10 folds higher after treatment of androgen and 5 folds higher after treatment of EGF than basal level in LNCaP, and 10 folds higher after treatment of EGF than basal level in DU-145. Conclusions Activation of MAPK may play a stronger role in the proliferation of androgen-independent prostate carcinoma cells than in androgen-dependent prostate carcinoma cells. Activation of MAPK may be involved in the proliferation of cells stimulated by androgen and EGF.
