摘要
抗人外周血T淋巴细胞单克隆抗体(McAb)SOKT_1通过SPDP与蓖麻蛋白A链(RTA)交联制成免毒素SOKT_1-RTA,在20mmol/L氯化铵存在条件下,能够杀伤82%人外周血T细胞,并使其~3H-亮氨酸掺入下降为对照的6.2%。SOKT_1-RTA的作用能够被未结合RTA的SOKT_1部分阻滞,而另一无关McAb末显示阻滞作用。表明:SOKT_1-RTA结合物具有SOKT_1的抗原结合特异性和RTA的细胞毒性,有可能成为清除人骨髓中T细胞的有效制剂。
An immunotoxin, SOKT_1-RTA, was prepared successfully by conju-gating a murine anti-human T lymphocytemonoclonal atibody (McAb) SOKT_1 andthe ricin toxin A chain (RTA) with theheterobifunctional linker SPDP. At thepresence of 20mmol/L NH.Cl, theSOKT_1-RTA conjugate killed 82% of thehuman peripheral blood T cells(PBT),and decreased their ~3H-Leu incorporationto 6.2% of the untreated controls. Thetoxic effect of the immunotoxin can bepertially blocked by unconjugated SOKT_1but not by an irrelevant McAb D_(?1). Theseresults indicated that the SOKT_1-RTAconjugate still retains both functions ofits component parts:binding to human PBTspecifically, and inhibiting protein syn-thesis. It may be an effective reagent forin vitro depletion of immunocompetentT cells from human bone marrows.
