摘要
本文报道采用Act-Ultrogel AcA 22亲和层析柱从胎儿尸体匀浆物内提取人体甲胎蛋白(αFP)的方法。此法费时短,收率较高,纯度和抗原性较好。提取的αFP用于^(125)I标记,可获得较好免疫活性的^(125)I-αFP,该标记品可用于αFP放射免疫分析。
In the present communica-tion, we report a method for the isolationand purification of αFP from fetal corpsehomogenate in a single-step procedure us-ing Act-Ultrogel AcA 22 affinity chromato-graphy column. This method proves to bebetter than the conventioal methodin simplicity, fastness and validity withrecovery, purity and immunoreactivity. Theimmunoeactivity of the purified αFP la-belled by the method of Hunter andGreenwood (1962) was 50-60% (B/T%). Thiswas similar to αFP standard (WHO) .Theresults indicate that the ^(125)I-αFP issuitable for αFP radioimmunoassay.
