摘要
目的介绍一种新的视网膜Müler细胞条件化原代培养的方法。方法使用ConA活化脾细胞培养上清液作为条件化培养基,分别培养人、大鼠、小鼠的视网膜Müler细胞,并进行形态学观察及免疫组织化学染色。结果用该方法原代培养不同种类的视网膜Müler细胞,免疫组织化学染色显示90%以上的视网膜Müler细胞的神经胶质纤维酸性蛋白(glialfibrilaryacidicprotein,GFAP)和S-100蛋白染色阳性。结论视网膜Müler细胞的条件化原代培养是一种可靠的、快速获得不同种类视网膜Müler细胞的理想方法。
ObjectiveTo introduce a method for conditioned primary culture of retinal Müller cells.MethodsConditioned medium was made of Con Aactivated spleen cell supernatant,and retinal Müller cells of human,rat and mouse were cultured,respectively.An identification was made with microscope observation and immunohistochemistry.ResultsWith the method,several kinds of retinal Müller cells were cultured,which showed positive reaction to glial fibrillary acidic protein (GFAP) and S100 protein with immunohistochemical assay.ConclusionConditioned primary culture of retinal Müller cells is a reliable and quick method.
