摘要
目的 :探讨川芎嗪对活动期狼疮肾炎 (LN)外周血单个核细胞 (PBMC)的B7- 1(CD80 )和B7- 2 (CD86 )mRNA表达的影响 ,为临床使用川芎嗪治疗LN提供理论依据。方法 :取活动期LN患者PBMC培养 ,分别以川芎嗪、地塞米松和川芎嗪 +地塞米松进行处理 ,以PBS对照 ,采用半定量反转录PCR的方法 ,检测PBMC的B7- 1和B7- 2mR NA表达。结果 :与对照组相比 ,川芎嗪组和地塞米松组均能明显抑制B7- 2mRNA表达 (P <0 .0 5 ) ,而川芎嗪 +地塞米松组对B7- 2mRNA表达的抑制更加明显 (P <0 .0 1)。各组对B7- 1mRNA表达均无显著影响 (P >0 .0 5 )。结论 :川芎嗪能下调活动期LN外周血单个核细胞B7- 2mRNA表达 ,合并使用地塞米松能更好地抑制B7- 2mRNA表达。
Objective: To investigate the regulation of ligustrazin on expression of B7-1(CD80)and B7-2 (CD86)mRNA in peripheral blood mononuclear cells (PBMC) from active stage lupus nephritis (LN), so as to obtain some principle evidences of ligustrazin treatment. Methods: PBMCs from active stage LN were cultivated and treated with PBS, ligustrazin, dexamethasone and ligustrazin plus dexamethasone injection respectively for 3 days. After cultivation, B7-1(CD80)and B7-2 (CD86)mRNA expressions were measured by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Results: Compared with PBS control group, both Ligustrazin and dexamethasone injection group check the expressions of B7-2 mRNA (P<0.05) and is more significant in Ligustrazin plus dexamethasone injection group. All group could not inhabit the expressions of B7-1 mRNA(P>0.05) Conclusion: Ligustrazin suppressed may significantly down-regulate expression of B7-2 mRNA in active stage LN and have synergetic effect with dexamethasone injection
