摘要
目的 :研究多药耐药基因 (mdr1)及 P-糖蛋白 (Pgp)单独及联合应用汉防己甲素 (Tet)和他莫西芬(Tmx)时 ,逆转多药耐药的效应。方法 :使用 MTT法对长春新碱 (VCR)、Tet、Tm x单独及联合使用时抑制 K5 6 2和 K5 6 2 / VCR细胞株的作用进行研究。结果 :当单独及联合使用 Tet和 Tmx时 ,它们不会增加 VCR对 K5 6 2细胞株的抑制率 ,但能明显逆转 K5 6 2 / VCR细胞株对 VCR的多药耐药。结论 :Tet和 Tmx单独使用均能逆转多药耐药 。
Objective:The classical multidrug resistance induced by multidrug resistance gene(mdr1)and p-glycoprotein(Pgp),is the major reason for the faliure of chemotherapy.When tetrandrine(Tet)and tamoxifen(Tmx)were used respectively and simultaneously,their effects of multidrug resistance reversal were studied.Methods:The effects of vincristine(VCR),Tet、Tmx used respectively and simultaneously on the inhibition of K562 and K562/VCR cells in vitro were studied by MTT.Rusults:When Tet and Tmx were used respectively and simultaneously,they will not increase the inhibition rate of VCR to K562 cells but can reverse the resistance of K562/VCR cells to VCR obviously.Conclusion:Tet and Tmx can reverse multidrug resistance respectively,when used together,they have distinct synergism.
