诱导胚胎干细胞向神经元分化及其在中枢神经系统损伤中的应用 被引量:5

摘要 胚胎干细胞是一种能在体外增殖 ,又能保持未分化状态的干细胞 ,在一定诱导下 ,胚胎干细胞可向多个方向分化 ,并生成多种功能细胞。近年来还发现在全反式维甲酸的诱导下 ,胚胎干细胞可生成神经元及神经胶质细胞等 ,这为利用胚胎干细胞进行中枢神经退行性变和损伤的治疗打下了基础。由于胚胎干细胞具有细胞株种类多、来源方便、诱导方法可靠易行等优点 ,因而在神经科学的各个领域有广阔的应用前景。本文对近年来胚胎干细胞体外诱导产生神经细胞以及在中枢神经系统损伤中的进展做一综述。 Embryonic stem cell (ES) is cloned stem cell line, which can either proliferate or keep undifferentiated in vitro. Under certain condition, ES cell is capable of differentiating into multiple cell types in vitro. Recently it is demonstrated that treated with retinoic acid (RA), ES cell can differentiate into glial cells (astrocytes and oligodendrocytes) as well as functional neurons. This provides a basis for using ES cell in therapy of degenerative diseases and injury of the central nervous system. Since many ES cell lines are available and the procedures for inducing differentiation of ES cells into neurons and glias are reliable and easy, the potential application of ES cells to neuroscience is very promising. In this paper we review the progress in ES cell induction in vitro and in the application of these cells to the central nervous system injury.
