A Comparative Study of Plasmid DNA Extractions

摘要 采用琼脂糖凝胶电脉对质粒 DNA 提取的三种方法进行比较,结果表明:煮沸法较为快速简便,提取质粒 DNA 含量较高;强碱法作用猛烈,质粒的共价、闭合、环状 DNA 构象容易破坏;SDS 裂解法作用较为温和,适合分子量大于10kbp 的质粒,大量提取时,煮沸法尤佳。 Using agarose gel electrophoresis,we compared three routine DNA extractions of plasmids through reagents and instruments of our nation.Our results showed that.①By lysis of boiling a lot of plasmid DNA could be extracted;②Lysis by alkali damaged CCC conformation of plasmid DNA by the combination of NaOH and SDS;③Lysis by SDS was suitable for extration of plasmids with size of more than 10kbp;④Lysis by boiling is favorable for large scale extraction of plasmid DNA.
