摘要
聚合酶链式反应仪 ( PCR仪 )是分子生物学的基本仪器。它通过设定不同温度变化条件对被扩增的 DNA片段进行变性、退火和聚合处理 ,以达到将 DNA片段的量成倍地扩增的目的。因此温度的分布和准确程度 ,尤其是各个温度的时间控制直接影响 DNA片段扩增的效率。通过定期对使用中的 PCR仪进行检测 ,可以了解仪器的工作状况 ,及时发现问题并加以维护、保养。
The polymerase chain reaction (PCR) machine is a basic instrument in molecule biology. In order to achieve the designed results of DNA amplification, the temperature variation distributed in the different wells of PCR block was checked. The results indicate that is should aware the temperature variation and adjust the set temperature setting according to PCR. By means of the tests of PCR machines in use on time we can realize the PCR's working condition and find problems and maintain it.
