丹参对转化生长因子β_1刺激的NIH/3T3细胞表达Ⅰ型胶原和c-fos mRNA的影响 被引量:9

摘要 目的:观察丹参对TGFβ_1刺激的NIH/3T3成纤维细胞功能的影响。方法:将丹参灌胃给正常大鼠,分离含药血清,温育TGFβ_1刺激的成纤维细胞。RT-PCR法检测Ⅰ型胶原和c-fos的基因表达。结果:经TGFβ_1刺激后,细胞Ⅰ型胶原和c-fos的mRNA表达明显增加,丹参含药血清可分别抑制由TGFβ_1引起的细胞Ⅰ型胶原和c-fos的mRNA表达的增加,结论:丹参可能通过抑制c-fos的基因表达进而抑制Ⅰ型胶原的基因表达。 Objective: To observe the effects of saliva miltiorrhiza on functions of transforming growth factor β1-stimulated fibrobiast. Methods: Serum of containing saliva miltiorrhiza was collected from rats and TGFβ1-stimulated fibrobiast cells incubated. Reverse transcription-polymerase chain reaction (RT-PCR) method was used to evaluate type Ⅰ collagen and c-fos mRNA expression. Results: After stimulated by TGFβ1, type Ⅰ collagen and c-fos mRNA expression obviously increased in fibrobiast. Drug serum of saliva miltiorrhiza could inhibit the increasing of type Ⅰcollagen and c-fos mRNA expression by TGFβ1-induced. Conclusion: saliva miltiorrhiza may be inhibited typeⅠ collagen mRNA . expression by suppressing c-fos mRNA expression.
