摘要
用紫外线照射诱发小鼠外周血淋巴细胞和肠淋巴结细胞的UDS(DNA周期外合成Unscheduled DNA Synthesis)银颗粒数随照射剂量的增加而增加,以20J/m^2为最佳;观察了小鼠经0.5、1、2Gy γ线全身照射后3天肠淋巴结细胞的修复合成,发现0.5Gy组无明显变化,而其余两个照射组细胞中的银颗粒数都低于正常;用此法测定了急性淋巴细胞白血病病人外周血淋巴细胞的UDS,此方法能清晰地识别S期合成与UDS,发现病人UDS并无变化,只是S期细胞高于同批健康对照。
The use of autoradiography to measure the unscheduled DNA synthesis (UDS) induced by 254 nm UV in lymphocytes from human blood and mouse mesenteric lymphadens wasdescribed. The results showed that the average grain count of mouse cell increased as theUV dose increased in the range of 5 to 40J/m2. The UDS induced by UV in mouse me-senteric lymphaden cells could be inhibited by gamma ray irradiation in vivo. Our data also suggested that the UDS was very similar in the blood lymphocytes from normal-individuals and from patients suffering from acute lymphocytic leukemia.
