摘要
本研究选用人类野生型SY5Y及SY5YA4CT基因转染细胞,应用免疫沉淀Western Blot等分析方法研究APP17肽对β-淀粉样肽及有关蛋白质如IDE、NT-3、JF表达的影响,探讨其影响神经元存活和退变的可能机理。结果发现,APP17肽能抑制转染细胞Aβ胞外分泌,并能促进野生型SY5Y细胞中IDE的表达,同时对内源性NT-3和NF的表达也有促进作用。提示:APP17肽抑制Aβ分泌和促进内源性神经营养因子的表达可能是其支持神经元存活、改善神经退变的机理之一。
Using wild type SY5Y cell lines and cell lines transfected with cDNA of APP C-terminal 100 aminal acids, we studied the effect of APP17-mer peptide on Aβ secretion and expression of related protein IDEsNT-3 and NF. The result indicated that APP17-mer peptide could increase expression of IDE.NT-3 and NF, but inhibit the secretion of Afi. The possible mechanisms underlying the protective effects of APP17-mer peptide on neurodegeneration are related to inhibiting Aβsecretion and increasing expression of endogenous neurotrophins.
