辐射诱导骨髓细胞凋亡及相关基因表达研究 被引量:9

Studies on hematopoietic cell apoptosis and the relative gene expression in irradiated mouse bone marrow

摘要 目的 研究辐射诱导骨髓细胞凋亡、bcl 2和 p5 3基因表达及意义。 方法 γ射线全身照射小鼠 ,采用原位末端标记、原位杂交和图像分析等技术 ,研究骨髓细胞凋亡及bcl 2和p5 3表达。结果 照射后 6h骨髓细胞凋亡 ;bcl 2于细胞凋亡时减少 ,而于其后修复早期增加 ;p5 3于造血细胞凋亡及之后修复均增多 ,而mtp5 3仅于其修复时阳性。 结论 辐射可导致骨髓细胞凋亡 ,且呈剂量相关性 ;bcl 2与 p5 3基因均参与其凋亡和修复过程。 Objective\ To study apoptosis and expressions of bcl\|2 and p53 in irradiated mouse bone marrow. Methods\ LACA mice were irradiated with 60 Co γ rays.By means of in situ terminal labeling, in situ hybridization and image analysis,the authors studied radiation\|induced apoptosis of hematopoietic cells and the expressions of bcl\|2 and p53. Results\ The characteristics of apoptosis appeared in hematopoietic cells at 6 hrs after irradiation.The expression of bcl\|2 was obviously decreased when apoptosis of hematopoietic cells occurred,whereas it increased in the early recovery phase;p53 protein increased during both apoptosis of hematopoietic cells and the recovery phase,and mutant type p53 DNA was positive only in the recovery phase. Conclusion\ Radiation may induced apoptosis of hematopoietic cells in a dose\|dependent manner;Both bcl\|2 and p53 genes play an important role in apoptosis and recovery phase.\;
