苦参碱对白血病细胞诱导分化作用和机理研究 被引量:29

摘要 为了解苦参碱对人粒系白血病细胞系HL 60细胞的诱导分化作用并探讨其机制 ,采用MTT法、光镜观察、S AP免疫组织化学法、硝基蓝四氮唑 (NBT)还原试验、流式细胞仪检测、逆转录酶 /多聚酶链反应 (RT PCR)检测 ,观察苦参碱对HL 60细胞的增殖抑制作用、分化诱导作用及其对细胞周期移行和癌基因表达的影响。结果表明 :苦参碱明显地抑制HL 60细胞的增殖 ,并诱导其向成熟粒系分化 ;苦参碱对HL 60细胞的诱导分化作用与其下调c myc基因表达、阻滞细胞在G1 In order to investigate the inducing and differentiating function of matrine on HL-60 cells and its mechanism,the proliferating and inhibiting function and the differentiating and inducing function of matrine on HL-60 cells were observed together with its influence on cell cycle migration and cancer gene expression,by MTT method,optical microscopy,S-AP immunohistochemical method,nitroblue tetrazolium(NBT)reduction test,and determination with flowing cell apparatus and reverse transcriptase/polymerase chain reaction(RT-PCR).The results showed that matrine could obviously inhibit the proliferation of HL-60 cells and induce them to differentiate toward the mature myeloid cells,and the inducing and differentating function of matrine on HL-60 cells might be related to its modulation of c-myc gene expression and retardation of cells in G 1 stage.
