摘要
目的从 ATP酶活性变化和自由基损伤方面研究老龄大鼠脑缺血再灌注心肌损伤机制。方法青年( 5月龄)和老龄( 20月龄以上)大鼠均分为模型组和正常对照组,观察大鼠全脑缺血 30分钟再灌注 60分钟后 ATP酶、乳酸脱氢酶( LDH)、肌酸磷酸激酶( CPK)、超氧化物岐化酶( SOD)的活性和丙二醛( MDA)含量。结果血清中 CPK和 LDH水平青年模型组明显高于青年对照组和老龄模型组,老龄模型组高于老龄对照组;老龄对照组血清中 CPK显著高于青年对照组。老龄模型组心肌 Na+- K+- ATP酶活性和青年对照组 Ca2+- ATP酶活性较老龄对照组和青年模型组均降低;老龄模型组心肌 Ca2+- ATP酶低于青年模型组和老龄对照组。老龄模型组血清 MDA/SOD比值高于老龄对照组,老龄模型组心肌组织 MDA/SOD比值高于青年模型组和老龄对照组。结论脑缺血再灌注心肌损伤可能与 ATP酶活性的变化和自由基损伤有关。但由于老龄大鼠 ATP酶活性和自由基代谢的增龄变化使脑缺血再灌注后这些病理改变较青年大鼠明显并具有一定特点。
Objective To study the mechanism of myocardial injury after brain ischemia reperfusion in aged rats by the changes of ATP ase and the injury of free radical.Methods Young (5 months) and aged (20 months or over) rats were divided into young control group (YCG),young model group (YMG),aged control group (ACG) and aged model group (AMG),the activitiy of ATP ase,lactic dehydrogenase (LDH),creatine phosphokinase (CPK) and superoxide dismutase (SOD),and the content of MDA were measured in the rats with 60 minute reperfusion after 30 minute brain ischemia.Results The serum CPK and LDH activities in the YMG were higher than that in the YCG and the AMG,and they were higher in the AMG than that in the ACG.The serum CPK activity in the ACG was higher than that in the YCG.The myocardial Na+ K+ ATP ase activity in the AMG and Ca2+ ATP ase activity in the YCG were lower than those in the ACG and the YMG;the myocardial Ca2+ ATP ase activity in the AMG was lower than that in the YMG and the ACG,the serum MDA/SOD ratio was higher in the AMG compared with the ACG,the myocardial MDA/SOD ratio in the AMG was obviously higher compared with that in the YMG and the ACG.Conclusions The myocardial injury after brain inchemia reperfusion in aged rats was correlated with the change of the activity of ATP ase and free radical injury.With aging,these pathological changes were obvious and distinctive compared with that in the young rats.
