不同剂量紫外线照射对浓缩血小板保存中细胞因子含量的影响 被引量:9

Effect of prestorage ultraviolet B radiation on the cytokine levels in stored platelet concentrates

摘要 目的 研究不同剂量B波段紫外线 (UVB)照射对浓缩血小板保存过程中细胞因子含量的影响。方法 采用富含血小板血浆 (PRP)法制备浓缩血小板 ,并分别以 1.5、2、2 .5J/cm2 UVB进行照射后 ,于标准条件下保存 5d ,间隔取样用ELISA方法检测IL 1β ,IL 6,IL 8及TNFα含量。 结果 对照组浓缩血小板在保存 1、3、5d后细胞因子含量与保存前比较差异有显著性意义 (P <0 .0 5 )。经 1.5J/cm2UVB照射后保存 3d细胞因子含量与保存前比较差异无显著性意义 (P >0 .0 5 ) ,而经 2J/cm2 、2 .5J/cm2UVB照射后 ,保存 5d细胞因子含量与保存前比较差异无显著性意义 (P >0 .0 5 )。各照射组中细胞因子含量在保存过程中明显低于对照组 (P <0 .0 5 )。结论 适当剂量的UVB照射能抑制浓缩血小板保存过程中细胞因子含量的升高 。 Objective To investigate the effect of prestorage ultraviolet B radiation on the cytokine levels during platelet concentrates storage. Methods Platelet concentrates prepared by the platelet rich plasma (PRP) methods,were irradiated by ultraviolet B at various doses, then stored to 5 days at standard condition. Cytokine levels were measured by ELISA at various storage time. Results The contents of cytokines in platelet concentrats of control group increased progressively with increasing storage time.Compared to prestorage,the irradiated unites of 1.5 J/cm 2 irradiation showed no significantly changes on 3 days storage (P> 0.05 ),the units of 2.0 J/cm 2 and 2.5 J/cm 2 showed no significantly changes on 5 days storage (P> 0.05 ).During storage,cytokine levels in irradiation groups were much lower than that in control group(P< 0.05 ). Conclusion Ultraviolet B radiation was effective in preventing the generation of cytokines during the storage of patelet concentrates.Thus Ultraviolet B irradiation at adequate dose can prevent non hemolytic febrile reaction after platelet concentrates transfusion.
