摘要
目的 研究趋化因子巨噬细胞炎性蛋白 2 (MIP 2 )在大鼠同种原位肝移植后的表达。方法 采用大鼠原位肝移植模型 ,将供肝用乳酸林格氏液保存 4h ,随机分为两组 ,即移植后 1h组和移植后 4h组 ,同时用假手术组作对照 ,对肝组织中的MIP 2mRNA的表达、血清MIP 2的表达、肝组织中的中性粒细胞浸润、血清丙氨酸转氨酶 (ALT)做了测定。结果 移植组MIP 2mRNA的表达和血清蛋白表达较假手术对照组显著升高 ,4h组高于 1h组 (P <0 .0 1) ,并且它的mRNA表达和血清蛋白表达与肝中中性粒细胞浸润、血清ALT变化呈正相关。结论 MIP 2在大鼠肝移植的缺血 /再灌注损伤中起重要的上调作用 ,成为肝移植再灌注损伤的重要因素之一。
Objective To study the expression of chemotactic factor macrophage inflammatory protein 2(MIP 2)following ischemia reperfusion injury of liver after rat orthotopic live transplantation. Methods By using rat orthotopic live transplantation model, donor liver was preserved in lactic acid Ringer's solution for 4?h (4?℃). The rats were then divided into two groups, namely, 1 h after transplantation and 4 h after transplantation. False operation group served as control. The expression of MIP 2 mRNA in liver and protein expression of MIP 2 in serum, neutropil infiltration in liver and ALT of serum were detected. Results In the donor liver pretreated with cold preservation for 4?h, compared with control group, the expression of MIP 2 mRNA and serum albumin in the group of 1 h after transplantation was increased significantly ( P < 0.01 ). The expression of MIP 2 mRNA and serum albumin in the group of 4 h was higher than that of 1 h group after transplantation ( P < 0.01 ). The expression of MIP 2 mRNA and albumin were positive correlation to neutrophil infiltration and ALT of serum with the correlation coefficient being 0.88 , 0.92 , 0.90 , 0.93 respectively ( P < 0.01 ). Conclusion MIP 2 might play an important upregulatory role in the ischemia reperfusion injury following rat orthotopic liver transplantation and become one of the most important factors in reperfusion injury of liver transplantation.
