利用紫外线和EMS诱变选育高产枯草芽孢杆菌 被引量:8

Breeding of UV and EMS-induced mutant strains of Bacillus subtilis with enhanced thrombolytic enzyme production

摘要 以豆豉溶栓酶产生菌株蜡状芽胞杆菌LD-8547为出发菌株,通过紫外线和甲基磺酸乙酯(EMS)的复合诱变选育高产枯草芽孢杆菌.用正交试验法优化EMS的诱变条件,得到最佳诱变条件,即在1 mL体系中,EMS用量20μL,诱变时间20min,菌液浓度1×108个·mL-1.最终经过初筛和复筛得到了1株产酶活性较高的菌株,对该菌株连续传代发酵培养5代,发现其产酶能力稳定,酶活力保持在16351 U·mL-1左右. Original strain (Bacillus subtilis LD-8547) was mutated by ultraviolet radiation and EMS induced mutation. Mutagcnesiscondition of EMS was optimized by orthogonal design, that was as follows: EMS was 20 IzL, bacteria liquid concentration was 1 ×10^8 mL, mutagenesis time was 20 win in a system of 1 mL. After two rounds of screening, a strain with high activity was selected.The enzyme activity of mutant was 16351 U·mL-1, which remained stable after five generations.
