摘要
目的探讨何首乌生品与炮制品游离蒽醌提取物的安全性,以最大耐受量给昆明种(KM)小鼠灌胃给药后动物产生的急性毒性反应,同时,评价其治疗剂量下对高脂血症大鼠肝肾功能的影响。方法采用单次给予小鼠最大耐受剂量的首乌游离蒽醌,观察给药不同时间点小鼠自主活动次数及14 d内体重变化、存活率。用高脂饲料喂养加灌服脂肪乳14d复制高脂血症大鼠模型,连续4周每日ig生(制)首乌游离蒽醌提取物40 mg·kg-1,模型组ig等量的生理盐水;停药1周后,检测血清中丙氨酸氨基转移酶(ALT)、天冬氨酸氨基转移酶(AST)及血清肌酐(CREA)。结果小鼠给药后未出现死亡情况,自主活动次数在观察时间内有明显变化(P<0.05);降低高脂血症大鼠因肝细胞损害而导致的ALT、AST升高。结论在本实验条件下首乌游离蒽醌LD50大于8000 mg·kg-1,制首乌能降低高脂血症大鼠AST到正常水平,未见明显的肝肾毒性损害。
Objective To discuss acute toxicity of processed products of Radix Polygoni Multiflori free anthraquinones extract to maximum tolerance to KM mice after intragastric administration of animal production, and further study of its effect on liver and kidney function in hyperlipidemie SD rats. Methods Radix anthraquinone in a given maximum tolerance dose of mice,the sponta- neous activity of mice were observed for 30 rain, 120 rain, 420 rain and 14 d body weight change, survival rate. By feeding high fat diet and fat emulsion duplicate the model of hyperlipidemia rats, for 2 weeks. After the model was made, were randomly divid- ed into four groups: normal group, rats model group, free anthraquinone (40 mg kg-I ) the treatment was administered for 4 weeks. Rats without any treatment in a week, detection of serum alanine aminotransferase ( ALT), aspartate aminotransferase (AST) and serum ereatinine (CREA). Results The free anthraquinone in Polygonum multiflorum in experimental acute toxicity in mice body weight in model group and control group had no significant difference ( P 〈 0.05 ). Radix Polygoni Muhiflori free anthraquinones extract treatment group can significantly reduce the content of hyperlipidemia rats serum of ALT, AST increased because of liver cell damage cause. Conclusion Under the condition of this experiment Shouwu free anthraquinone in LDs0 more than 8000 mg~ kg-l. The processing Polygonum multiflorum can reduce hyperlipidemia AST rats to normal levels, liver and kid- ney toxicity was not found.
