摘要
采用丹磺酰氯作为衍生剂,建立鱼制品中4种生物胺(腐胺、尸胺、组胺、酪胺)同时检测的高效液相色谱分析方法,对售常见新鲜鱼类及制品中生物胺的含量进行检测。首先将样品进行柱前衍生,后经C18(4.6×150 mm×5μm)柱,柱温30℃,及最佳洗脱条件下将样品中不同的生物胺分离并定量。新鲜鱼在室温下经不同时间贮藏后分别测定其肉中生物胺的积累情况,结果表明随时间的延长,鱼肉内生物胺的含量急剧增加,24 h后生物胺的总量最高能达到2096.5μg/g。鱼类制品的分析结果表明,各样品中生物胺的含量较均匀,且多数保持在较低水平。鱼类产品中,生物胺的含量与其新鲜程度及产品品质相关,控制好生物胺的产生是保证产品品质与质量的关键。
Methods of simultaneous determination of four biogenic amines (putrese ine, cadavefine, histamine, tyramine) by high-performance liquid chromatography with a pre-column dansyl chloride derivation procedure were developed. Biogenic amines in several fish products were determined. First, the samples were pre-column derivatization, then using C18 (4.6 × 150 mm×5 μm) column, column temperature 30 ℃, and gradient eluting under optimal conditions, so different biogenic amine in the sample were separated and quantitated. The accumulation of biogenic amines in fish has been determined after stored in different period time. The content of biogenic amines in fish has increased sharply with the storage time increased. The maximum amount of biogenic amines could reach 2096.5/ag/g after 24 h of storage. The result of determination of fermentation fish products showed that the content of biogenic amines was stable and most of them kept a lower level. The freshness of fish products related the content of biogenic amines. Keeping the producing of biogenic amines is important for fish products' quality.
