摘要 目的:探讨维生素K2对SCA3/MJD的神经保护作用。方法:构建Gmr-GAL4/UAS系统和Nrv2-GAL4/UAS系统两种不同表达部位的SCA3/MJD转基因果蝇模型。并分别用1.00,0.10和0.01mM的维生素K2干预。结果:0.10mM的维生素K2明显抑制了SCA3/MJD转基因果蝇眼睛视网膜光感受神经元变性,降低果蝇翅膀异常率,改善果蝇运动能力,0.01mM的维生素K2作用较0.10mM的弱,1.00mM的维生素K2反而加重SCA3/MJD转基因果蝇眼睛视网膜光感受神经元变性,增加果蝇翅膀异常率,降低果蝇运动能力。结论:维生素K2对SCA3/MJD转基因果蝇的神经保护作用与维生素K2剂量有关,过低作用弱,而过高损伤神经元。 Objective: To investigate the neuroprotective effect of vitamin K2 on SCA3/MJD. Methods: To construct Gmr-GAL4/UAS and Nrv2-GAL4/UAS systems, which expressed at different parts in SCA3/MJD transgenic Drosophila models. SCA3/MJD transgenic Drosophila models were interfered with 1.00, 0. 10 and 0.01 mM. Vitamin K2. Results: 0.10 mM vitamin K2 significantly inhibited SCA3/MJD transgenic Drosophila eye retinal photoreception neuronal degeneration, reduced the abnormal rate of Drosophila wings, improved exercise capacity, the effect of 0.01 mM vi- tamin K2 was weaker than that of 0.10 mM vitamin K2, 1.00 mM of vitamin K2 increased SCA3/ MJD transgenic Drosophila eye retinal photoreception neuronal degeneration, increased anomaly rate of Drosophila wings, reduced the locomotor ability. Conclusion: The neuralprotective effect of vitamin K2 on SCA3/MJD transgenic Drosophila is associated with vitamin K2 dose, too low, weak role, and too much, damage to neurons.
