myc-cyclin B1重组基因的克隆表达和细胞内定位

摘要 细胞周期蛋白B1(cyclin B1)是调控细胞由G2期向M期转换的重要因子之一, G2末期cyclin B1的表达达到高峰。激活细胞周期蛋白依赖性激酶2(cyclin dependent kinase 2, CDK2), 形成cyclin B1/CDK2复合物, 进入细胞核, 裂解一系列底物蛋白, 推动细胞从DNA合成期进入有丝分裂。肿瘤细胞的永生化与细胞周期的调控失常有密切关系, 并且发现, 作为一种细胞周期调节因子, cyclin B1与肿瘤有着非常密切的关系, 它是与肿瘤有直接关系的癌基因之一[1-2]。Cyclin B1[3]在许多癌细胞中均存在高表达[4-5], 如在卵巢癌、胃肠癌、宫颈癌和膀胱移行细胞癌中。在细胞周期G2/M起正性调控作用, 启动有丝分裂, cyclin B1高表达使细胞周期紊乱, 促进肿瘤的发生[6-7]。使用抗癌药可以降低cyclin B1表达, 细胞周期阻滞在G2/M期[8]。本实验拟构建cyclin B1的真核表达载体, 并验证其在细胞内的定位情况, 为进一步研究cyclin B1与肿瘤的关系奠定了实验基础。
