摘要
目的:探讨灯盏花素对2型糖尿病(T2DM)大鼠急性心肌缺血模型的保护作用及机制。方法:高脂高糖饮食联合链脲佐菌素(STZ)制备T2DM大鼠模型,将成模大鼠随机分为模型组、灯盏花素低(100mg/kg)、高(200mg/kg)剂量组,并设正常对照组。给药14d,末次给药1h后除正常组外各组皮下注射异丙肾上腺素(ISO)6mg/kg,3d造心肌缺血模型。末次给ISO后记录大鼠麻醉状态下Ⅱ导联心电图,取心脏组织行HE染色,取血测空腹血糖、空腹胰岛素并计算胰岛素抵抗指数、总胆固醇、甘油三酯、高密度脂蛋白-胆固醇、低密度脂蛋白-胆固醇水平,ELISA法测定血清脂联素(APN)含量,免疫组化法检测心肌组织APN表达。结果:灯盏花素100mg/kg、200mg/kg可减少Ⅱ导联心电图ST段偏移,降低空腹血糖、空腹胰岛素、胰岛素抵抗指数、总胆固醇、甘油三酯、低密度脂蛋白-胆固醇水平,减轻心肌组织病理损害,上调APN的表达,其中200mg/kg组比100mg/kg组效果佳。结论:灯盏花素对T2DM大鼠急性心肌缺血模型具有明显的保护作用,其机制可能与调节糖脂代谢、改善胰岛素抵抗和低脂联素血症有关。
Objective: To study the protective effect and mechanism of breviscapine on type 2 diabetes mellitus( T2DM) rats with acute myocardial ischemia. Methods: The T2DM model rats were induced by feeding on high-fat and high-carbohydrate diet and injecting streptozotocin and then were randomly divided into the model group,the low dose group( 100mg /kg),and the high dose group( 200mg /kg). A normal control group was also set up. The medication was given for 14 days. One hour after the last drug delivery,Myocardial ischemia injury models were established by subcutaneous injection of ISO( 6mg /kg,continuous 3d) except the normal control group. After the last administration,the changes of ECG( Ⅱ) in anesthetic conditions were recorded,the myocardial tissue was obtained and stained with hematoxylin and eosin,fasting blood glucose( FBG),insulin( INS) and such items were calculated as the insulin resistance index( HOMA-IR),total cholesterol( TC),triglycerides( TG),high-density lipoprotein cholesterol( HDL-C),and low-density lipoprotein cholesterol( LDL-C) level were measured.The serum level of adiponectin( APN) were measured by ELISA. The expressions of APN were analyzed by immunohistochemistry. Results:Breviscapine( 100 and 200mg /kg) can reduce ECG LeadⅡ st-segment deviation and decrease the level of FBG,INS,( HOMA-IR),TC,TG and LDL-C. It can also lessen pathological injuries of the myocardial tissues,increase adiponectin levels( P < 0. 05). Curative effect of the200mg / kg group was better than the 100mg /kg group( P < 0. 01). Conclusion: Breviscapine has certain preventive and therapeutic effect on myocardial ischemia in rats with type 2 diabetes mellitus,which may have a dependence on the dosage. The mechanism may be related to regulation of glucolipid metabolism,improvement of the insulin resistance and hypoadiponectinemia.
