摘要
目的观察黄芩茎叶总黄酮对Ⅱ型胶原诱导性关节炎小鼠脾淋巴细胞Th1、Th2及其相关细胞因子IL-10、IFN-γ的影响,初步探讨黄芩茎叶总黄酮对类风湿性关节炎的作用。方法选用C57BL/6小鼠,建立鸡Ⅱ型胶原诱导性关节炎小鼠模型,将造模成功的小鼠随机分为CIA模型组、黄芩茎叶总黄酮组、雷公藤多苷组,另设正常对照组,于初次免疫后第21天开始灌胃给药,35 d后流式细胞术检测小鼠脾淋巴细胞Th1、Th2水平及相关细胞因子IL-10、IFN-γ的表达。结果与模型组比较,药物组小鼠CD4+T淋巴细胞中Th1细胞数量明显降低(P<0.05),Th2比例增加(P<0.01),药物组IL-10分泌显著增多(P<0.05),IFN-γ分泌明显减少(P<0.05),2用药组无明显差异(P>0.05)。结论黄芩茎叶总黄酮可以调节Th1/Th2的平衡及其相关细胞因子IFN-γ、IL-10的表达。
This study designed to explore the therapeutic effect of the scutellaria baicalensis stem-leaf total flavonoid (SSTF) on rheumatoid arthritis model by observation the effect of SSTF on the Th1 and Th2 cells and related cytokines IL-10, IFN- γ C57BL/6 mice were chosen as experimental objects, and randomly divided into collagen-induced arthritis(CIA) model group, scutellaria group, triptolide group, and normal control group. For drug groups, the drug was administered 21 d after the first immunization, once a day for 35 days; while mice in the normal control group and model group were administered saline. The CD4+ T lymphocyte subsets of Th1 and Th2 and the related cytokines IL-10, IFN-γ were detected with flow cytometry technique. Drug groups demonstrated higher levels of Th 1 and IL-10 (P〈 0.05), but lower levels of Th2 (P〈 0.01) and IFN- γ (P〈 0.05), as compared with the model group. While no significant difference was found between the 2 drug groups (P〉 0.05). All result indicated that SSTF can balance the CD4+ T lymphocyte subsets Th1/Th2 cells and the related cytokines IL-10 and IFN-γ.
