摘要
目的:观察巴马汀对脂多糖(LPS)诱导的RAW264.7小鼠巨噬细胞一氧化氮(NO)的分泌及诱导型一氧化氮合酶(iNOS)表达的作用。方法:采用LPS诱导RAW264.7巨噬细胞建立细胞炎症反应模型,加入不同浓度的巴马汀,采用Griess法测定NO的分泌量;采用Western blot法检测iNOS蛋白的表达。结果:巴马汀能明显抑制LPS诱导的RAW264.7巨噬细胞NO的分泌,以及iNOS蛋白的表达,并呈现出浓度依赖性。结论:巴马汀可通过抑制LPS诱导的RAW264.7巨噬细胞NO的分泌及iNOS的表达,从而发挥抗炎作用。
Objective: To investigate the effect of palmatine on nitric oxide (NO) production and inducible nitric oxide syn- thase (iNOS) expression induced by lipopolysaccharide (LPS) in RAW264. 7 mice macrophage cells. Methods : An inflammatory re- sponse model was established using RAW264. 7 macrophage cells induced by LPS. Dose of paimatine were used to treat RAW264.7 macrophage cells to evaluate the effects of palmatine. NO concentration was measured with Griess reagent and iNOS expression was test- ed by Western blot. Results: Palmatine markedly inhibited the secretion of NO and iNOS protein expreesion in LPS-stimulated RAW264.7 macrophage cells in a dose-dependent manner. Conclusion: The anti-inflammatory effects of palmatine may be mediated by inhibiting the secretion of NO and the expression of iNOS protein.
