一氧化氮——一氧化氮合酶系统对卵巢切除鼠骨丢失的影响 被引量:4

EFFECT OF NITRIC OXIDE-NITRICOXIDE SYNTHASE SYSTEM ON BONE LOSS IN OVARIECTOMIZED RATS

摘要 目的 观察一氧化氮 (NO)供体硝酸异山梨酯 (IsosorbrideDinitrate,ID)及一氧化氮合酶 (NOS)抑制剂氨基胍 (Aminoguani dine ,AG)对双侧卵巢切除 (OVX)大鼠股骨的作用。方法  49只雌性大鼠随机分成 7组 ,每组 7只。①假手术 (Sham)组、②卵巢切除(OVX)组、③卵巢切除 +雌激素 (OVX +E)组、④卵巢切除 +NO供体 (OVX +ID)组、⑤卵巢切除 +NO供体和雌激素 (OVX +ID +E)组、⑥卵巢切除 +NOS抑制剂 (OVX +AG)组、⑦卵巢切除 +NOS抑制剂和雌激素 (OVX +AG +E)组。实验 1 0周后处死大鼠 ,测定大鼠股骨的重量和骨密度 (BMD) ;并测定血清NO、NOS水平。结果 与Sham组相比 ,OVX组、OVX +AG组、OVX +AG +E组大鼠股骨重量减轻、BMD减少 (P <0 .0 5) ;而OVX +E组、OVX +ID组、OVX +ID +E组与Sham组之间无明显差异 (P >0 .0 5)。结论 NO供体预防OVX鼠骨丢失 。 Objective To investigate the response of femur bone of ovariectomized (OVX) ratsto nitric oxide (NO) donor isosorbride dinitrate (ID) and nitric oxide synthase (NOS) inhibitor aminoguanidine )AG). Methods 49 Mistar female rats were randomly divided into 7 groups (7 in each group): 1) sham-operated (Sham); 2)OVX; 3)OVX treated with estrogen (OVX+E); 4) OVX treated with NO donor (OVX+ID); 5)OVX treated with NO donor plus estrogen (OVX+ID+E); 6)OVX treated with NOS inhibitor (OVX+AG); 7) OVX treated with NOS inhibitor plus estrogen (OVX+AG+E).At the end of 10 weeks of the experiment, the rats were killed and their femoral weight, bone mineral density (BMD) and their serum NO and NOS levels were measured. Results The data suggest that the weight and BMD of femur were remarkably reduced in OVX group, OVX+AG group and OVX +AG+E group (P<0.05), while they were similar in OVX+E group, OVX+ID group and OVX+ID+E group compared with the Sham group (P>0.05). Conclusion NO donor has the preventive effect of bone loss of PVX rats and NOS inhibitor can block the estrogen to prevent the OVX rats from bone loss.
