摘要
目的:通过建立T淋巴细胞(T细胞)免疫功能缺陷的小鼠动物模型,观察壮尔颗粒对小鼠体内细胞因子平衡状态的影响。方法:将48只小鼠随机分为空白组,模型组,玉屏风颗粒组,壮尔颗粒高、中、低剂量组,空白组和模型组ig给予生理盐水,其余各组分别ig给予按玉屏风颗粒溶液(3.38 g·kg-1),壮尔颗粒高、中、低剂量组(4.04,2.02,1.01 g·kg-1)。给药10 d后,空白组经鼻滴入10μL无菌生理盐水,其余各组经鼻滴入10μL肺炎链球菌悬液(密度为1×105CFU/mL),制备小鼠T细胞免疫功能缺陷的动物模型。应用流式微球技术(CBA)分别测定小鼠血清和小鼠脾脏白介素(IL)-2等细胞因子的水平,免疫组织化学染色法检测小鼠脾脏中IL-2和IL-4的表达。结果:与正常组比较,模型组小鼠血清和脾脏中IL-2,肿瘤坏死因子(TNF),干扰素-γ(IFN-γ)的水平均有显著性下降(P<0.05),同时IL-4,IL-6,IL-10,IL-17A的水平有显著性升高(P<0.05)。与模型组比较,壮尔颗粒高、中剂量组可显著提高小鼠血清和脾脏中IL-2,TNF,IFN-γ水平(P<0.05),同时IL-4,IL-6,IL-10,IL-17A的水平均有显著性下降(P<0.05),并且小鼠脾脏IL-2表达上调和IL-4表达下调,有统计学意义(P<0.05)。结论:壮尔颗粒能够通过调节小鼠体内细胞因子的平衡来增强机体免疫功能。
Objective: To study effect of Zhuang'er granules on adjusting cytokines balance in T lympboeytes (T-cell) immunodeficiency mice. Method: Forty eight mice were randomly divided into six groups: normal group, model group, Jade screen granules group, high-dose Zhuang'er granules group, middle-dose Zhuang'er granules group and low-dose Zhuang'er granules group, normal group and model group received NS ig respectively. Meanwhile, other groups received Jade screen granules (3.38 g·kg-1), high-dose Zhuang'er granules (4.04 g·kg-l), middle-dose Zhuang'er granules (2.02 g· kg-1) and low-dose Zhuang'er granules (1.01 g ·kg-1) ig respectively. After ten days, the mice in normal group were infected with 10 μL of NS, other groups were infected with 10 μL of streptococcous pneumoniae. The T-cell immunodepression mice models were induced by injection of cyclosporine A. The general symptoms of animals were observed. The levels of interleukin (IL) -2, tumor necrosis factor (TNF), interferon-γ (IFN-γ), IL-4, IL-6, IL-10, IL-17A in the serum and spleen were detected by cytometric beads array, IL-2, IL-4 were detected by immunohistochemistry. Result: Compared with the normal group, the levels of IL-2, TNF, IFN-γ decreased (P 〈 0.05) in the serum and spleen of model group, the levels of IL-4, IL-6, IL-10, IL-17A increased (P 〈 0.05). Compared with the model group, the high and middle dosage of Zhuang'er granules group can improve the levels of IL-2, TNF, IFN-γ in the serum and spleen (P 〈0.05) , the levels of IL-4, IL-6, IL-10,IL-17A decreasing (P 〈0.05). Compared with the model group, there was no difference in lower dosage of Zhuang'er Granules group; IL-2 expression was increased, IL-4 expression was decreased (P 〈 0.05 ). Conclusion: Zhuang'er granules can improve immunodeficiency and immunological function of mice by Adjusting Cytokines balance.
