摘要
目的:考察姜黄素白微乳在Beagle犬体内的血药浓度经时过程并评价其相对生物利用度。方法:Beagle犬分别单剂量给予姜黄素自微乳及其混悬液,通过HPLC测定血浆中姜黄素的质量浓度,流动相甲醇(A)-水(B)梯度洗脱(0-15min,30%~70%A;15~30min,70%~90%A),检测波长425nm,采用DAS2.1.1程序拟合药-时曲线,计算药代动力学参数及生物利用度。结果:姜黄素自微乳与混悬液的药代动力学参数t_max分别为3.0,2.0h,C_max分别为3304.3,1873.6ng·L-1,AUC_0.24依次为6537.5,2973.2ng·h·L-1,姜黄素自微乳相对于混悬液的生物利用度达219.88%。结论:姜黄素自微乳可显著提高药物在Beagle犬体内的生物利用度。
Objective: To investigate plasma concentration-time process of curcumin self-microemulsions in Beagle dogs and evaluate its relative bioavailability. Method: Beagle dogs were given a single dose of curcumin microemulsions and its suspensions, HPLC was adopted to determine the concentration of curcumin in plasma with mobile phase of methanol (A) -water (B) (0-15 min, 30%-70% A; 15-30 min, 70%-90% A) for gradient elution and detection wavelength at 425 nm, pharmacokinetics parameters and bioavailability were calculated by DAS 2. 1.1 programs. Result: T_max of curcumin self-microemulsions and suspensions were 3.0 and 2.0 h, C_max were 3 304.3 and 1 873.6 ng·L-1 , AUCo_24h were 6 537.5 and 2 973.2 ng ·h ·L-1 of curcumin self-microemulsions relative to curcumin suspensions were 219.88% microemulsions could significantly improve bioavailability in Beagle dogs. , respectively. Conclusion : Bioavailability Curcumin self-microemulsions could significantly improve bioavailability in Beagle dogs.
