HPLC法测定抗病毒栓中的鸟苷,(R,S)告依春,百秋里醇和广藿香酮 被引量:4

摘要 目的采用高效液相梯度洗脱法定量测定抗病毒栓(板蓝根、广藿香、石膏、知母等)中鸟苷、(R,S)-告依春、百秋里醇和广藿香酮。方法Hypersil C18色谱柱(4.6mm×250mm,5μm);体积流量0.8mL/min。鸟苷和(R,S)-告依春以甲醇-0.5%磷酸溶液为流动相梯度洗脱,检测波长253nm。百秋里醇和广藿香酮以乙腈-0.2%磷酸溶液为流动相梯度洗脱,检测波长308nm。结果鸟苷和(R,S)-告依春分别在0.0218—0.4360μg(r:0.9994)、0.0836~1.672μg(r=0.9992)范围内进样量与峰面积呈良好的线性关系,平均加样回收率分别为96.8%、97.5%,RSD(n=6)分别为1.1%、1.3%;百秋里醇和广藿香酮分别在0.2328~4.656μg(r=0.9997)、0.0132~0.2640μg(r=0.9995)范围内进样量与峰面积呈良好的线性关系,平均加样回收率分别为98.1%、96.8%,RSD(n=6)分别为1.4%、1.1%。结论该方法测定结果准确、灵敏、重复性好。 AIM To develop an HPLC method for determining the content of guanosine, (R, S)-goitrin, pat- choulic alcohol and pogostone in Kangbingdu Suppositories (Isatidis Radix, Pogosternonis Herba, Gypsum fibrosum, Anemarrhenae Rhizoma, etc. ). METHODS An Hypersil Cls column was used as the chromatographic column, the flow rate was 0. 8 mL/min. Guanosine and (R,S)-goitrin were determined in a gradient elution manner using methanol-0. 5% phosphoric acid as the mobile phase and the UV detection wavelength was set at 253 nm. Patchoulic alcohol and pogostone were determined in a gradient elution manner using acetonitrile-0. 2% phosphoric acid and the UV detection wavelength was set at 308 nm. RESULTS There was a good linear relationship between the concentration of guanosine, (R, S)-goitrin and peak area value as the concentrations of guanosine and (R, S)-goitrin were within the ranges of 0. 021 8 -0. 436 0μg (r =0. 999 4), 0. 083 6 - 1.672μg (r =0. 999 2), respectively. The average recoveries were 96. 8% ( RSD = 1.1% ) and 97.5% ( RSD = 1.3% ), respectively. There was a good linear rela- tionship between the concentration of patchoulic alcohol, pogostone and peak area value as the concentrations of patchoulic alcohol and pogostone were within the ranges of 0. 232 8 -4. 656 Ixg (r =0. 999 7), 0. 013 2 -0. 264 0 μg (r =0. 999 5) , respectively. The average recoveries were 98.1% (RSD = 1.4% ) and 96. 8% (RSD = 1.1% ) , respectively. CONCLUSION The method is accurate, sensitive, reproducible and can be used in the determi- nation of guanosine, (R, S)-goitrin, patchoulic alcohol and pogostone in Kangbingdu Suppositories.
