胰岛素促进犬在体心肌细胞葡萄糖转运子4基因表达 被引量:3

摘要 目的探索胰岛素促进心肌细胞葡萄糖摄取增加的机制。方法采用 Northern法分析心肌 GL U T4m RNA和免疫法分析心肌 GL U T4多肽。结果胰岛素刺激心肌 GL UT4m RNA和 GL UT4多肽表达增加 1~ 1.2倍。同时伴随心肌葡萄糖摄取增多。结论胰岛素能刺激 GL UT4m RNA和 GL U T4多肽表达 ,使 GL UT4数增加 ,进而促进心肌葡萄糖摄取增多 ,胰岛素刺激心肌细胞 GL U T4表达 ,可能是心肌增加葡萄糖摄取的重要分子学机制之一。 ObjectiveTo investigate the mechanism for that insulin facilitates increase of glucose uptake MethodsThe expression of myocardial GLUT4 polypeptide was determined by semiquantitative immunoblotting The expression of GLUT4 mRNA was determined by semiquantitative Northern blotting ResultsAfter infusing insulin for 8 hours,the expression of GLUT4 mRNA and GLUT4 polypeptide was significantly higher in canine myocardium than in those found normal ones The glucose uptake was upregulated at the same time.ConclusionsOur findings suggest that insulin facilitates the expression of GLUT4 mRNA and GLUT4 polypeptide in canine hearts Enhanced GLUT4 expression is one of the important molecular mechanism by which myocardial cells enhance glucose uptake by insulin stimulation
