摘要
目的 :探讨人胎脑低分子肿瘤抑制物对白血病细胞的作用机制及其细胞内游离Ca2 +浓度 ([Ca2 +]i)的变化。方法 :通过超滤技术分段提取胎脑低分子抑瘤物 ,采用光镜、相差显微镜和流式细胞仪检测凋亡 ,Fura 2荧光负荷技术测 [Ca2 +]i。结果 :胎脑低分子抑瘤物可明显抑制K 5 6 2白血病细胞的增殖 ,作用 4h后可出现典型的凋亡改变。白血病细胞凋亡过程中 ,胞内 [Ca2 +]i明显升高。结论 :胎脑低分子抑瘤物可能是通过诱导白血病细胞凋亡而抑制其增殖 ,在白血病细胞凋亡过程中 [Ca2 +]i升高。
Objective: To study the effects of a low molecular weight tumor supppressor (LMW TS) from human fetal brain on leukemia cells and changes in intracellular free calcium levels ([Ca 2+ ]i). Methods: LMW TS was obtained through ultrafiltration; its inhibitive effect measured with MTT, apoptosis investigated by means of light microscopy and flow cytometry; and [Ca 2+ ]i determined by Fura 2 fluorescin load technic. Results:LMW TS from human fetal brain could obviously inhibit K 562 cells, after treatment with 2 μg/ml LMW TS for 4 hours, typical apoptosis appeared, and [Ca 2+ ]i significantly increased in the initial stage. Conclusion : LMW TS from human fetal brain may suppress the leukemia cells through the access of apoptosis induction, and [Ca 2+ ]i level increased in the process of leukemia cell apoptosis.
