摘要
[目的]建立用Folin-Ciocaileu法测定红腺忍冬叶抗氧化有效部位中总多酚含量的方法,并应用此方法研究不同批次红腺忍冬叶抗氧化有效部位中总多酚含量的变化。[方法]采用Folin-Ciocaileu法,对红腺忍冬叶水提液中总多酚含量测定进行方法学研究,检测12批红腺忍冬叶水提液及其30%乙醇沉淀物和滤液中总多酚的含量。[结果]测得总多酚含量在6μg·mL-1~16μg·mL-1范围内具有良好的线性关系(r=0.9996),重复性RSD为0.31%(n=6),平均加样回收率为98.67%,RSD为2.66%(n=9)。不同批次红腺忍冬叶间测得总多酚含量差别较大;同批次红腺忍冬叶中,总多酚含量为水提液〉水提液30%乙醇沉淀后滤液〉水提液30%乙醇沉淀。[结论]该方法简单、准确、重复性好,可用于测定红腺忍冬叶抗氧化有效部位中总多酚的含量。红腺忍冬叶抗氧化有效部位中总多酚含量为水提液〉水提液30%乙醇沉淀后滤液〉水提液30%乙醇沉淀。
[Objective] To establish a Folin-Ciocaileu method for determination of the contents of total hydroxybenzene in antioxidant effective fraction of leaves of Lonicera Hypoglauca Miq. which is used in the exploration of the content changes of total hydroxybenzene in antioxidant effective fraction of leaves of Lonicera Hypoglauca Miq. of different batches. [Methods]The methodology of the method for determination of the contents of total hydroxybenzene in water extraction of leaves of Lonicera Hypoglauca Miq. was investigated by Folin-Ciocaileu method. The contents of total hydroxybenzene in water extractions or the precipitation and filtrate of 30%ethanol precipitating action from 12 batches of leaves of Lonicera Hypoglauca Miq. were determined.[Results] The methodological study showed that the linear range of total hydroxybenzene was 6 μg·mL^-1~16 μg·mL^-1 (r=0.9996). The repetitiveness RSD of total polyphenol was 0.31%(n=6). In addition, the average recovery of total polyphenol was 98.67%, with RSD 2.66%(n=9). The total hydroxybenzene contents in different batches extraction of leaves of Lonicera Hypoglauca Miq. were quite different. While the contents of total hydroxybenzene in the same leaves of Lonicera Hypoglauca Miq. were: water extraction>the filtrate of 30% ethanol precipitating action>the precipitation of 30% ethanol precipitating action.[Conclusions]The method is simple, accurate, reproducible and suitable for simultaneous determination of the contents of total polyphenol in antioxidant effective fraction of leaves of Lonicera Hypoglauca Miq..
